What is Tamoxifen?

Tamoxifen is a hormonal therapy medication used to treat and prevent breast cancer. It is classified as a Selective Estrogen Receptor Modulator (SERM). Unlike chemotherapy, which kills cells directly, Tamoxifen works by blocking the hormones that certain types of cancer need to survive and grow.


Mechanism of Action

The drug is specifically designed for Hormone Receptor-Positive (HR+) cancers. In these cases, cancer cells have receptors that act like “locks,” and the hormone estrogen acts like a “key” that tells the cells to multiply.

Tamoxifen acts as a “decoy key.” It enters the lock (the receptor) but doesn’t turn it. By occupying that space, it prevents real estrogen from attaching to the cancer cells, effectively starving them of the signals they need to grow.


Pharmacological Profile

Tamoxifen is unique because its behavior changes depending on where it is in the body:

  • In the Breast: It acts as an anti-estrogen, blocking cell growth.

  • In the Bones and Uterus: It acts as an estrogen agonist (meaning it mimics estrogen). This can be beneficial for maintaining bone density, but it is also why the medication requires careful monitoring for its effects on the uterine lining.


Clinical Application

Tamoxifen is generally administered as a daily oral tablet. It is used in several clinical stages:

  • Adjuvant Therapy: Taken after primary treatments (like surgery) to prevent the cancer from returning.

  • Metastatic Treatment: Used to slow or stop the spread of advanced breast cancer.

  • Prophylaxis: Given to individuals at a statistically high risk of developing breast cancer to lower their chances of a future diagnosis.
